The line was built for the Midland Railway and the station which stood between Castle Gresley and Church Gresley opened on 2 October 1848. It was originally sited to the west of the road bridge and around 1869 it was repositioned to the east of the bridge. Gresley station closed on 7 September 1964; however, the line remains open for freight traffic. In later years there have been plans to restore passenger services between Leicester and Burton including reopening a station at Gresley, which would also serve the nearby town of Swadlincote.
Between 1906 and 1927 a branch of the Burton and Ashby Light Railway terminated at the station. This was a tramway system operating in three counties from Burton-upon-Trent, Staffordshire to Swadlincote in Derbyshire and then to Ashby-de-la-Zouch in Leicestershire.
